Gene : NSI A. thaliana

DB identifier  ? AT1G32070 Secondary Identifier  ? locus:2195361
Name  ? nuclear shuttle interacting Brief Description  nuclear shuttle interacting
TAIR Computational Description  nuclear shuttle interacting;(source:Araport11)
TAIR Curator Summary  Encodes a acetyltransferase (NSI) that is localized in the nucleus and chloroplast. It interacts with the geminivirus movement protein NSP. This interaction is required for viral infection and systemic spread. Acetylates the viral coat protein (CP) in vitro, but not NSP. NSP inhibits NSI activity in vitro. In the chloroplast NSI functions in the dynamic reorganization thylakoid membrane complexes. NSI is highly transcribed in phloem and in xylem parenchyma cells, and in the apical meristem and guard cells, within young tissues in Arabidopsis, and its expression is turned off as tissues mature.Mutants have reduced melatonin and anthocyanin levels and do not accumulate the PSI-LHCII state transition complex. The protein has distinct lysine acetylation and relaxed N-terminal acetylation specificities on chloroplast proteins as determined by in vitro as well as in vivo analyses using quantitative protein mass spectrometry (PMID:32633465).

Type Comment Proteins
GNAT2_ARATH-2 GNAT2_ARATH GNAT2_ARATH-3 developmental stage Expressed in the apical meristem, root vasculature and all veins in the cotyledons of young developing seedlings (PubMed:16461385). Loss of expression in the older maturing tissues (PubMed:16461385). Accumulates transiently in developing seeds (e.g. including embryos and seed coat), reaching a peak ten days after pollination (10 DAP) (PubMed:32354877).
GNAT2_ARATH-2 GNAT2_ARATH GNAT2_ARATH-3 function (Microbial infection) Required for begomovirus infection and systemic spread (PubMed:12837950, PubMed:15452236, PubMed:16461385). In case of begomoviruses infection, acetylates the capsid protein (CP), but not the nuclear shuttle protein (NSP) (PubMed:12837950, PubMed:15452236, PubMed:16461385). Stimulates melatonin-triggered defense responses to the necrotrophic Botrytis cinerea (PubMed:34234798).
GNAT2_ARATH-2 GNAT2_ARATH GNAT2_ARATH-3 function Protein acetyltransferase with dual specificity triggering both N-alpha-acetylation (NTA), with a preference for alanine, serine, threonine, methionine and to a lower extent valine as substrates (can also use glycine and leucine), and epsilon-lysine acetylation (KA) of several plastid proteins (PubMed:32633465). Triggers lysine acetylation in KEA1 and KEA2 (PubMed:29967049, PubMed:31865509). Acetylates in vitro histones H2A and H3 (PubMed:12837950). Does not act as a transcriptional activator but required for the dynamic reorganization of thylakoid protein complexes and grana during photosynthetic state transitions (PubMed:16461385, PubMed:29967049, PubMed:35792507). Involved in melatonin biosynthesis by catalyzing the formation of N-acetylserotonin (NAS) from serotonin and of melatonin (N-acetyl-5-methoxytryptamine) from 5-methoxytryptamine (5-MT) (PubMed:25250906, PubMed:32354877). By triggering melatonin biosynthesis, contributes to the chloroplast protein quality control (CPQC), which plays a pivotal role in starch synthesis, and confers melatonin-associated tolerance to high light (HL) stress (PubMed:29770489, PubMed:33806011). Prevents the accumulation of oil and anthocyanin content in mature seeds and avoids seed germination in a melatonin-dependent manner, but promotes mucilage production in the seed coat (PubMed:32354877, PubMed:33811388). Contributes to melatonin-mediated anthocyanin production in cold-exposed seedlings (PubMed:27047496). Implicated in melatonin-monitored circadian dynamics of stomatal aperture to minimize night water loss and promote drought tolerance, partly by triggering hydrogen sulfide H(2)S-dependent stomotal closure in response to osmotic stress (PubMed:32064655, PubMed:33924609, PubMed:34812898).
GNAT2_ARATH-2 GNAT2_ARATH GNAT2_ARATH-3 similarity Belongs to the acetyltransferase family. GNAT subfamily.
GNAT2_ARATH-2 GNAT2_ARATH GNAT2_ARATH-3 tissue specificity Highly expressed in cauline leaves and seeds, at lower levels in stems, siliques, inflorescences and rosettes leaves and at very low levels in roots (PubMed:12837950, PubMed:16461385, PubMed:31698875, PubMed:32354877, PubMed:32633465). Expressed in the xylem parenchyma and phloem of the leaves and root, and in guard cells of young leaves (PubMed:12837950, PubMed:16461385, PubMed:32633465).
