Description: | The aim of the experiment is to find out the contribution of the modification of the host-plant transcript profiling to the generation of a phenotype as a result of the transformation.The whole roots were used for the isolation of RNA.Transgenic and wild type plants were cultivated in hydroponics medium for 6 weeks. Three independent experiments were performed.In each experiment, roots from eight plants grown in different containers were pooled and frozen in liquid nitrogen.RNA was isolated with RNeasy Plant Mini Kit, Quiagen. RNA was treated with the RNAse-free DNAse (Promega).There were prepared three samples from the control material (wild-type tobacco), and three samples from transformed tobacco - grown in three independent experiments. Samples were sent on dry ice for analysis. |
