Title:A genomic approach to understanding seed dormancy

Description:Background: Physiological seed dormancy is present throughout the higher plants and has a profound impact on many aspects of crop production and the conservation of genetic resources as well as the structure and development of plant communities. Despite this, there is considerable ignorance of how dormancy is controlled at the molecular level. Greater understanding of seed dormancy therefore has considerable generic value. Aims: The long-term goal is improved understanding of seed dormancy and the identification of generic molecular markers of seed dormancy status. The specific aim of the applications was to compare gene expression patterns in Arabidopsis Cvi seeds in 13 physiological states (52 arrays) Information on selected A. thaliana ecotype: The test organism is Arabidopsis thaliana accession "Cape Verde Islands" (Cvi). Characterisation shows that mature, primary dormant seeds (at shedding) require a dry storage treatment (after-ripening) of up 6 months to induce a uniform non-dormant state in the seed population. When imbibed, germination will then ensue if the seeds are provided with appropriate conditions of light, nitrate or low temperature. Alternatively, if a non-dormant seed is imbibed in the dark they enter a state of secondary dormancy in which a moist-chilling treatment is required to re-induce a state of non-dormancy. Experimental design: A full experimental design and description of the treatments used is given in the two publications below. Table 1 in each publication summarises the 13 treatments.
