Title:Measurement of pre-existing and newly transcribed mRNA at two ambient temperatures

Description:In Arabidopsis thaliana a number of genes show altered transcript abundances at different ambient temperatures. As with other biochemical reactions, mRNA transcription and decay rates might be expected to increase passively with increasing temperature but it is not clear to what degree active temperature responses play a role in temperature-regulated genome-wide gene expression changes.Using a non-perturbing base analogue (4-thiouridine) labelling and separation method we have measured the relative abundances of both labelled, newly transcribed transcripts and unlabelled, pre-existing transcripts. Together with unseparated samples and samples taken at the start of the labelling time, these arrays have been used to calculate transcription and decay rates of the Arabidopsis thaliana transcriptome at two ambient temperatures.
