Description: | The objective of this experiment is to proof the concept of tetrapyrrole-mediated signaling by analysis of the primary response on nuclear transcription by using an inducible transgene expression system. Transgenic plants with chemical induced silencing (RNAi) of genes encoding enzymes of the Mg branch (chlH, chlM, chl27) will allow us to explore time-resolved early variations of nuclear gene expression upon induced changes of activities of these specific steps in the chlorophyll-synthesizing branch of tetrapyrrole biosynthesis.For transcriptome analysis we used green tissue of three weeks old arabidopsis seedlings treated with dexamethasone for 24 h. The seedlings were grown in little pots on soil under the same conditions. |
