Title:Comparative gene expression analysis between Arabidopsis wt and wrky33 mutant plants upon Botrytis c

Description:Wrky33 mutant plants are highly susceptible to the fungus Botrytis cinerea (Bc) while wt is not. To identify differentially expressed genes 5 weeks old wt and wrky33 (GABI_324B11) plants were mock treated and spray infected with Bc (isolate 2100 Spanish Type Culture Collection) at 5x105 spores/ml, and kept for 14 hours at high humidity and constant light. To resuspend the spores and for mock treatment Vogel buffer was used (in 1l: 15g sucrose, 3g Na-citrate, 5g K2HPO4, 0.2g MgSO4 7H2O, 0.1g CaCl2 2H2O, 2 g NH4NO3). 100 mg of leaf tissue was collected and total RNA isolated using TRI reagent following the suppliers instructions (Ambion). Leaves from three plants were pooled to make one biological replicate, and samples from three independent biological replicates for each genotype and treatment were submitted to hybridisation against a total number of 12 Arabidopsis ATH1 GeneChip arrays (Affymetrix).
