Title:Response to gamma-irradiation in Arabidopsis thaliana mutant and wild-type seedlings

Description:Background: At5g24280 is an ATM dependent gamma-irradiation inducible gene (Culligan et al., The Plant Journal, 2006). To evaluate its possible role in the transcriptional response upon ionizing radiation, we collected total RNA from wildtype and one At5g24280 T-DNA insertion line upon gamma-irradiation. Experimental Setup: Seeds were sterilized, washed and imbibed in ddH2O for 3 days at 4°C, sown on 1/2 MS plates and grown under continuous light at 22°C for 5 days. Plants were irradiated using a Cs137 source (100Gy), returned back into the growth cabinet and harvested 1.5h post irraditaion. RNA was extracted from irradiated and control seedlings using Tri-Reagent (Sigma), then DNase-treated with TurboDNase (Ambion) following the manufacturers suggestions. Additional purification with PCI /CI extraction was followed by EtOH-precipitation. Strains: wildtype plants and the At5g24280 T-DNA insertion line (SALK_036931) are in a Columbia (Col-O) background.
