Title:Cis-jasmone as a novel signalling molecule in plant gene expression

Description:(Z)-Jasmone (cis-jasmone) is a highly volatile product of jasmonic acid (JA) catabolism and is not only a constitutive compound of flower volatiles but also induced by insect damage of vegetative tissue. It has been speculated that cis-jasmone might be a biologically inactive sink for the JA pathway but recently it was demonstrated that cis-jasmone acts as an insect semiochemical. In addition, we have shown by differential display that cis-jasmone induces plant gene expression qualitatively distinct from that elicited by methyl jasmonic acid (Birkett et al, PNAS 97: 9329-9334). Thus it is likely that cis-jasmone is a new lipid-derived signalling compound as opposed to the previously proposed inert sink.We are therefore interested in using the Arabidopsis DNA microarray to search for genes whose expression is altered by cis-jasmone treatment. As it is not known yet what role cis-jasmone might play in oxylipin signalling, we hope that the microarray identification of such genes will give an indication as to whether cis-jasmone is involved solely in defence signalling or whether it has additional functions.Arabidopsis thaliana ecotype Columbia will be exposed to cis-jasmone for 1 or 3 hours respectively. For each treatment (and the mock treated control plants) 8 rosettes will be harvested, pooled and used for RNA extraction. Three biological replicates will be generated per treatment.
