Title:Cold Acclimation Time Course

Description:OBJECTIVE: To determine expression profile of Arabidopsis (Columbia) under temperature stress and to identify genes that may be involved in short-term and long-term stress tolerance.Temperature stress treatments consist of three sub experiments each beginning at 2 hr into the light period:1- heat shock at 40C over 4 hr with same light intensity as control2- cold shock at 4C over 96 hr with same light intensity as control3- deacclimation after cold acclimation for 96 hr at 20C for 24 hr with same light/dark conditions.Note: In these experiments circadian clock regulation is taken in to account as much as possible with 2 possible controls taken.1- Control (20C) was taken 2 hr after the lights turned on and this was the time temperatures stress samples were taken2- control (20C) 6 hr after the lights turned on and this was the time that 4 hr heat stress and 4 hr cold stress samples were takenPlant Growth Conditions:Light: Photosynthetically active radiation is 100-150 micromol/m2/s. Light regime is 15/9 hr light/dark cycles. Cool white fluorescent and incandescent lamp are used as light sources. Light quality was very uniform throughout the growth area at all temperature conditions.Soil: Plants were grown in Fafard 2 Mix (Canadian Sphagnum Peat (55%), Perlite, and Vermiculite). Soil is autoclaved.Temperature: Plants were grown at 20C for about 3 weeks (8.5 leaf stage) at 15/9 D/L with irradiance as stated above prior to initiation of temperature treatments.All aerial tissue above the soil line was harvested for RNA an sample. Each sample was composed of 16 plants.Kaplan et al., Plant Physiol. 136, 4159-4168.
