Title:Using transcriptome analysis to define the developmental state of fn10, a mutant in Arabidopsis thal
Description:The fn10 mutant was identified from the same high-dormancy mutant screen as the cts-1 (comatose) mutant (Russell et al. 2000; Footitt et al. 2002). As well as showing reduced germination potential, the fn10 mutant is hypersensitive to abscisic acid (ABA) and fails to establish on sucrose.In these experiments the transcriptome profile of fn10 seed that has been stored for 1 month will be compared with after-ripened fn10 seed that has been stored for 6 months. Under optimal conditions the non-after-ripened fn10 seed will not germinate, whereas the after-ripened seed will. We will also look at after-ripened fn10 seeds imbibed on 100 nM ABA, a concentration that will prevent germination of the seed. Control samples will be after-ripened wild-type Landsberg erecta (Ler) seeds, and Ler imbibed on 10 ?M ABA, a concentration that will prevent germination.From these experiments, and from experiments previously performed by the group, we hope to be able to define the developmental state of non-germinating fn10 as either dormant or non-dormant. We hope this will aid us in the understanding of this mutant, and the role the protein plays in the control of dormancy and germination.Reference List 1. Footitt S. et al.
